Epithalon is a synthetic tetrapeptide that produces the same effect as epithalamin, a natural polypeptide produced by pineal gland. It was first developed by Professor Vladimir Khavinson, a Russian scientist, in 1980. Its main function is to enhance the production of telomerase, an enzyme that helps cell produce telomers.
Telomers promote the stability and integrity of chromosomes by preventing the loss of essential DNA sequences during cell division. But as people age, telomere length shortens, and the progressive shortening of telomers affects the health and lifespan of humans. Further, it is involved in DNA regulation, and melatonin production and shows significant effects against cancer and some infectious diseases.

Epithalon and Antiaging Different scientific studies indicate that epitalon can prolong the lifespan and delay the aging process. A study found that epitalon preparation when added in the nutrient medium at the developmental stage can increase the mean life span of adult Drosophila Mongoster by 11-16% [1]. Another research on Swiss-derived SHR mice concluded that this tetrapeptide slows down the age-related declining estrous function and can lessen the occurrence of chromosomal abnormalities in bone marrow [2].Similarly, one in vitro experiment found that epitalon can slow down the aging process of oocytes. Furthermore, it reduced the level of ROS (free radicals that can damage healthy cells) in aged oocytes, which explains a possible reason for its antiaging effect. [3]. Apart from omitting free radicals, it is involved in the activation/production of telomerase, as discussed above. One in vitro study on human somatic cells confirmed the telomerase-inducing activity of epithalon. Further, it suggested the possible role of epitalon in prolonging lifespan in the individual cell and the whole organism [4].
Epithalon and Cancer Epithalon has been studied for its antitumor activity. Several studies show that epitalon tetrapeptide can stop and prevent the development of tumors in rats. Further, it suggested that downregulation of HER-2/neu gene expression might be responsible for the anticancer activity of this peptide[5, 6]. In a study, epitalon reduced the development of mammary tumors in HEN/u2 mice and decreased the size of lung metastases [7]. epitalon has also been shown to stop the initiation of breast carcinogenesis in these mice. An article suggests that epitalon, along with epithalamine, might be useful in the prevention of breast cancer in women who are at risk [8].
Epitalon and Skin Health One in vitro study involving the organotypic cultures of tissues from the skin of animals suggests that epitalon can be used in cosmetics to strengthen and restore the skin structure. This effect can be explained based on its antioxidant and telomerase-inducing activity, which slows down the aging of the skin and improve overall appearance [9]. Furthermore, one article suggests that it suppresses caspase-dependent apoptosis (decreases caspase activity responsible for programmed cell death) that increases during the process of aging [10].
Epitalon and Cardiovascular Effects In a 12-year randomized clinical study, patients treated with epithalamine showed improved functional cardiovascular health and increased exercise tolerance. Plus the cardiac mortality was twice less in patients treated with peptide than those in the control group.[11]. Moreover, another clinical study demonstrated that epithalmine improved lipid metabolism and arterial blood pressure in women with ischemic heart disease and hypertension [12].
